He served as the 2nd assistant director for the 2002 Hindi-language period romantic drama film Devdas, starring Shah Rukh Khan, Aishwarya Rai, and Madhuri Dixit.
In 2008, Sikander Kher made his acting debut as Sameer Chopra/Akshay Kapadia in the Hindi-language mystery thriller film Woodstock Villa. He co-starred in the film alongside Gulshan Grover, Arbaaz Khan, Sachin Khedekar, and Neha Oberoi.
He played the recurring role of Haroon Sherchan (the younger brother of Roshan Sherchan who wants his brother to be out of prison) in the Hindi-language action thriller television series 24 in 2016. The show is based on the American series of the same name.
Sikander Kher brought to life the character of Rudra in an episode of the American science fiction drama streaming television series Sense8 in 2017.
